. Tobramycin, also known as topramycin, is an aminoglycoside antibiotic. Aminoglycoside antibiotics have no conjugated structure and weak UV absorption, so they are not suitable for routine direct analysis using a UV detector. In the 2020 edition of the Chinese Pharmacopoeia, related substances in tobramycin raw materials are detected using an evaporative light scattering detector (ELSD). Compared with CAD, ELSD has lower sensitivity and poorer repeatability; the assay item uses a microbiological method with relatively poor specificity. In current USP standards, related substances of tobramycin are detected by thin-layer chromatography, which cannot provide in-depth analysis of individual impurities; the assay method uses UV detection after sample derivatization and is relatively cumbersome. Therefore, in a draft for comment issued by the United States Pharmacopeial Convention in January 2023 [1], an HPLC-CAD method was adopted for detecting related substances and assay of tobramycin.
