Tobramycin, also known as Topramycin, is an aminoglycoside antibiotic. Aminoglycoside antibiotics lack conjugated structures and have weak UV absorption, making them unsuitable for direct analysis using conventional UV detector methods. The 2020 edition of the Chinese Pharmacopoeia uses an evaporative light scattering detector (ELSD) for the determination of related substances in tobramycin raw material. Compared with CAD, ELSD has lower sensitivity and poor repeatability; the content determination uses a microbiological assay method, which has poor specificity. The current USP standard uses thin-layer chromatography for the determination of related substances in tobramycin, which cannot provide in-depth analysis of each impurity; the content determination method involves sample derivatization followed by UV detection, which is cumbersome. Therefore, the United States Pharmacopeial Convention issued a draft for comment in January 2023 [1], proposing an HPLC-CAD method for the determination of related substances and content of tobramycin.
