2′-Fucosyllactose (2′-FL) is an oligosaccharide abundant in human milk and belongs to human milk oligosaccharides (HMOs). It supports probiotic growth, inhibition of harmful bacteria and immune regulation.

2′-FL has been recognized by the U.S. FDA as Generally Recognized as Safe (GRAS) and is used as a food ingredient in infant formula.

HPLC-CAD, high-performance liquid chromatography coupled with Charged Aerosol Detection, is suitable for qualitative and quantitative analysis of 2′-FL because of its high sensitivity and wide linear range. It has been used for structurally related impurity analysis in 2′-FL purity reference materials developed by the National Institute of Metrology, China and a dairy company in Heilongjiang, and for value assignment of 99.2% 2′-FL and 89.5% LNnT reference materials.

Instruments: Agilent 1200 HPLC system + Instrumax CADetector a1 Charged Aerosol Detector.

Reagents: acetonitrile (chromatographic grade), pure water (Watson’s distilled water). Samples: lactose, galactose and 2′-Fucosyllactose (2′-FL).

2. Methods and results

2.1 CAD conditions

Instrument model: Instrumax CADetector a1. Drift tube temperature: 40 °C. Nebulizing gas flow: 3 L/min. Charging current: 1 μA. Charging gas flow: 1 L/min.

HPLC-CAD is an efficient detection method with high value for 2′-FL research, development and quality control. As the application potential of 2′-FL in food nutrition continues to be explored, HPLC-CAD is expected to play an even more important role in this field.
